چکیده

During last decade, proteomics has emerged as a powerful tool for high throughput screening, identification and quantification of proteins from biological samples. In current study, we have employed proteomics technique to explore proteins from eye lenses of Uromastyx hardwickii, an Indian spiny-tailed lizard. Water soluble and insoluble proteins from Uromastyx hardwickii lens were separated and characterized by 2-DE (two dimensional electrophoresis). After ingel digestion, well resolved spots were identified by nanoliquid chromatography-electrospray tandem mass spectrometry (nLC-MS/MS). We have identified αA-, βA2-, βA4-, βB2and βB3-crystallins as ubiquitous crystallins and δ-, τ-crystallins as taxon-specific lens crystallins in water soluble fraction. αAand βB3-crystallins were found to be most abundant water soluble proteins in Uromastyx hardwickii lens. Among non-crystallins (cellular proteins), β-actin was identified. From water insoluble fraction, vimentin, gelsolin, gamma enolase like protein, filensin like protein and ATP synthase subunit B were identified. Our results illustrate advantage of proteomics technology for identification of lens proteins. Furthermore, the study provides distribution patterns of ubiquitous and taxon specific crystallins from Uromastyx hardwickii that is likely to be useful in understanding evolutionary lineage of this organism.

Lactate dehydrogenase like crystallin: a potentially protective shield for indian spiny-tailed lizard (Uromastyx hardwickii) lens against environmental stress?

Taxon specific lens crystallins in vertebrates are either similar or identical with various metabolic enzymes. These bifunctional crystallins serve as structural protein in lens along with their catalytic role. In the present study, we have partially purified and characterized lens crystallin from Indian spiny-tailed lizard (Uromastyx hardwickii). We have found lactate dehydrogenase (LDH) activ...

Seasonal variations on serum glucose and lipid profile in a male poikilotherm vertebrate, Uromastyx hardwickii.

BACKGROUND Uromastyx hardwickii, a local lacertilian species, is widely distributed in the Indo-Pak subcontinent and inhabits mostly the arid regions of the country. This study was carried out to assess the seasonal variations on glucose, cholesterol and triacylglycerol (TAG) levels in a poikilotherm vertebrate species and to develop an experimental model for the study of lipid metabolism in hi...

Comparison of optic lens proteins among animals at different stages of development

The purpose of this investigation was to study and compare the electrophoretic patterns of optic lensproteins of different species of domestic animals at pre- and post-natal ages. Optic lenses were removed from the embryo or adult sheep, cattle, goat, camel and chicken at the slaughter-house then homogenized and subjected to sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE). I...

The C terminus of lens aquaporin 0 interacts with the cytoskeletal proteins filensin and CP49.

PURPOSE Aquaporin 0 (AQP0), the most abundant membrane protein in the lens, is a water-permeable channel, has a role in fiber cell adhesion, and is essential for fiber cell structure and organization. The purpose of this study was to identify proteins that interact with the C terminus of AQP0, by using a proteomics approach, and thus further elucidate the role of AQP0 in the human lens. METHO...
